Short description of project
Diabetes mellitus is one of ten leading underlying causes of mortality in Macau. Patients with diabetic mellitus
suffer from complications including diabetic foot ulcers, which can result in lower limb amputations. The
methyltransferase SET7/9 interacts and methylates HIF-1α, triggering the breakdown of HIF-1α in the nucleus.
Elevation of SET7/9-mediated methylation and consequent destabilization of HIF-1α induced by hyperglycemia
contributes to the pathophysiological mechanisms of diabetic wounds. Thus, stabilization of HIF-1α by blocking
the interaction of SET7/9 and HIF-1α could be a potential strategy to improve wound healing, especially in diabetes.
In this study, we aim to identify inhibitors of the SET7/9–HIF-1α interaction from libraries of bioactive compounds or in-house metal-based compounds for the treatment of diabetic wounds.
